Runs perfectly with PROTON_USE_WINED3D=1. Otherwise I got a blank screen and all inputs were captured.
CTD after latest patch
Most recent patch completely broke proton compatibility. Game CTDs while loading a saved game or starting a new one and no tinkering seems to fix this issue.
Spawns a black window with blue borders. Sound works fine, but there is no video. Tweaks either do nothing or prevent even the window from spawning.
As per everyone else, you need to install dotnet40, and everything is fine. This game also runs in VR, though you will need strong hardware for a smooth experience. With a 1080 Ti and an HTC vive, I'm getting a lot of stutters, though the game is playable this way.
Runs perfectly fine out of the box, but a good graphics card is recommended. The game runs at 50-55 fps in 4k, which is slower than I expected. At 1920x1080, it's perfect.

Run with PROTON_FORCE_LARGE_ADDRESS_AWARE=1 or it will seem to crash to loading screen like others have noted. The game doesn't seem to fully close after running, so you may need to do a pkill -9 -f Kynseed after quitting.
No changes needed at all. Performance is a little rough. I'm usually staying above 60 fps at 1920x1080, but it should be a lot better with my hardware.
Some performance issues, but nothing problematic. Loading takes longer than on windows and framerate struggles to stay at 60 Hz on a 4k display with maxed settings a lot of times, but it's hardly an issue.
Runs perfectly, though force feedback does not function. The framerate in VR is fine, but the game will only output to your headset while you're in the car. You'll need to set the game up, then quickly put on your headset.
The instructions everyone's following didn't quite work for me. A few changes were needed to get this to work for me. Set sound to alsa and bring up winecfg as usual, but I needed to set my Z drive to ~/.local/share/Steam or else steam was unable to locate the game exe. The standard changes to Rocksmith.ini were needed. Audio was horribly garbled until I made changes to /etc/pulse/daemon.conf: default-fragments = 5 and default-fragment-size-msec = 2. Additionally, I couldn't see the graphic to tune my guitar until disabled d3d11. After all is said and done, the game runs fine, now. There's minor input lag, but it's hardly a show-stopper.
This is an update to an earlier report where I claimed gold status. This game has grown increasingly unstable with time. First I'd sometimes need to start the game twice due to strange graphical glitches. Then it became a '2-3 times is the charm' affair. Then 4-5. Still, once you got it going, it was normally okay. Now the game sometimes crashes aftera few songs even if everything seems to be perfect. This title appears to be regressing.
The game will not output to my headset. From my flatscreen, I can see that it is otherwise tracking and working perfectly, though.
KDE Plasma panels appear on top of the game in borderless window mode. Switching to full screen fixes this problem entirely, and the game behaves when alt-tabbing.
Framerate is fine on high detail. Some weird graphical glitches, but nothing game breaking. As others have mentioned, your mouse will go nuts after going into a menu. Alt-tab only fixed this after I ran with -force-opengl. I also lose my mouse pointer in menus a lot. Alt-enter fixes that.
Title screen is messed up. Otherwise, it plays fine. This game will not stream and will cause steam to crash if you try.
I've tried disabling esync and d3d11 as well as using wine d3d11, but nothing allows this game to launch. I don't even get a window.
