It's working out of the box, don't touch the prefix, leave it on the default, because it could corrupt the game.

The game doesn't start.
I tried to change to every major proton release, but none of it helped. The default, Proton 7, Proron 6 and Proton 4. Borked.

When I first launched the game I had the black screen issued mentioned by other users. I launched the game again and it worked perfectly.
Unknown(< 15 minutes with Proton)Sometimes the configuration screen doesn't come up. But after that it was possible to play the game. You may have to create ~/.local/share/Steam/SteamApps/compatdata/302080/pfx/drive_c/users/steamuser/My Documents/Bus Driver/config.cfg by your own to make it work. My res of 1920x1080 was not supported by the game. But setting unsupported resolutions like mine caused it to have rendering issues (e.g. disappearing lines for designated stops).

Unknown(2+ hours with Proton)Use first the Setup Tool to set the screen resolution, and check the active GPU first. The game runs fine with an Intel GPU, if you reduce the resolution to improve performance. To use the NVidia card on switchable graphics laptops, use optirun %command% on startup shortcut to use the high performance GPU instead.

Once done, all settings are update, and the game will run without great issues.
